GIPZ Lentiviral shRNAmir triggers for RNAi combines superior shRNAmir design with the advantages of the GIPZ lentiviral vector to produce a powerful tool for RNAi studies. The pGIPZ lentiviral vector was designed based on the PRIME series of vectors developed by the Elledge Lab. shRNAmir constructs targeting the entire human genome have been cloned into the pGIPZ lentiviral vector to produce the Human GIPZ lentiviral shRNAmir library. Transduction efficiency of pGIPZ

   TurboGFP expression from the pGIPZ vector allows measurment of transfection and transduction efficiency. It also effectively marks shRNAmir expression in target cells. A. HEK293 cells 48 hrs post-transduction and B. the corrosponding phase image. 100% transduction efficiency is seen in these cells
Knockdown efficiency of GIPZ lentiviral shRNAmir


HEK293T cells were transduced with lentiviral particles containing shRNAmir to GAPDH, eG5, p53, Rb or STAT6. non-silencing shRNAmir was transduced as a control. The graph depicts the residual levels of these genes normalized to their equivalent non-silencing control

The GIPZ lentiviral shRNAmir library is provided in 96-well microtiter plates containing frozen stock cultures of E.coli  in LB (low salt) broth with 8% glycerol, carbenicillin (100ug/ml) and zeocin (25ug/ml).  Individual shRNAmir constructs are shipped as glycerol stock cultures on wet ice. Open Biosystems checks all cultures for growth prior to shipment. Store at -80ºC.
