Fluorescent Arrest-In

Dual function reagent for shRNA delivery
Fluorescent (Fl) Arrest-In™ is sulforhodamine B-conjugated Arrest-In™. It was developed to enable visualization in real-time, of Arrest-In-DNA complexes in cells. Fl Arrest-In™ is a  dual function reagent, permitting visualization of transfected complexes as well as efficient delivery of shRNA into the nucleus of transfected cells. Features include:
  • Lipo-polymer based formulation optimized for high efficiency delivery of shRNA
  • Rhodamine fluorescent tag enables visualization and tracking of transfection events
  • Perform double label experiments
  • Works effectively on adherent and suspension cell lines
  • Low cytotoxicity
Double label studies with Green Fluorescent Protein (GFP)
Figure 2. Hela cells stably expressing GFP were transfected with Fl Arrest-In-DNA complexes. Images were generated 5 and 48 hours after transfection. Red: Fluorescent Arrest-In-DNA complexes
Green: GFP expression Blue: Nucleus Yellow: Co-localized Fluorescent Arrest-In and GFP
The rhodamine fluorescent tag (excitation is at 560nm and emission at 580nm) allows double label experiments with a variety of commonly available tags (GFP, FITC, biotin etc.). Fl Arrest-In has been tested on a variety of cell types and has shown transfection efficiencies comparable to that seen using unconjugated Arrest-In™. Studies to assay for cytotoxic effects of both Arrest-In and Fl Arrest-In show that both reagents had minimal to no cytotoxic effects when compared against untreated cells. See product insert for data.
