Drosophila RNAi library-DNA templates

High throughput RNAi screening
The Expression Arrest™ Drosophila RNAi Library-dsDNA templates is a collection of over 15,000 dsDNA templates ready for in vitro transcription (IVT) to dsRNA constructs that target the entire Drosophila genome.  dsDNA templates were generated using gene specific primers to amplify 200-800 bases of exonic sequence for each gene. Features include:
  • dsDNA templates for targeting the entire Drosophila genome
  • Perform high-throughput RNAi screening in Drosophila cells
  • Dual T7 promoters for direct use in in vitro transcription
  • Sufficient material to generate over 600ugs of dsRNA 

    The Drosophila RNAi Library 1.0 and 2.0 collectively target the entire Drosophila genome with dsDNA templates ready for generation of dsRNA constructs for RNAi. The dsDNA templates can successfully generate microgram quantities of dsRNA in a single IVT reaction. Using five randomly picked constructs from our collection with two commercially available kits, IVT reactions were performed. The RNA obtained ranged between 21 and 67 µg, using as little as 1ul of the template. The variation in yield is both template and kit dependent. With the 10µl of dsDNA provided, over 600 µg of RNA can be generated. The variation in yield is both template and kit dependent. Most protocols that involve microinjections, cell culture or transfection of RNAi in Drosophila require 0.1µg to 30µg of dsRNA. Download the pdf for a copy of the complete experiment.

Shipping Information:
Drosophila RNAi Library 1.0 and 2.0 are available in a high-throughput format in 96-well PCR plates. Plates contain frozen constructs and are shipped on dry ice. Store at –20° C. Individual constructs (available from Drosophila RNAi library 1.0 only) are shipped on wet ice.
