an Open Labs Product for gene functional analysis of an intracellular pathogen

Created by a consortium of laboratories1, the Brucella ORF Collection is comprised of 3091 ORFs that have been cloned into a Gateway® entry vector and are ready to be transferred into Gateway destination vectors for protein expression and functional analysis (Figure 1). For example, protein-protein interactions can be studied by transferring the collection to bait and prey plasmids for yeast two-hybrid screening or to a destination construct that expresses each ORF with an affinity, epitope, or fluorescent tag.

Shipping Information:
Brucella ORF clones are available as glycerol stocks. Individual clones are shipped in microtubes at room temperature, while the entire collection is shipped in 96-well plates on dry ice. Microtubes and plates should be stored at –80ºC.

C. jejuni ORF Collection
 

Jodi Parrish et al1 from the Center for Molecular Medicine and Genetics, and Department of Biochemistry and Molecular Biology, Wayne State University have cloned 1,619 ORFs from Campylobacter jejuni into E. coli using in vivo recombination. The ORFs that were chosen include 1654 ORFs that were predicted by the Sanger Institute2 , together with 31 additional ORFs predicted by TIGR (http://www.tigr.org/tigr-scripts/CMR2/CMRHomePage.spl). The availability of these ORFs circumvents a common rate-limiting step in proteomics, the generation of an appropriate predicated ORF clone set for use in proteome-scale micro-array analysis. Their use in micro-array studies to profile gene expression patterns will be invaluable to the research community and will provide a quick and inexpensive way to investigate genes and proteins and understand their function and regulation.

E. coli SURE clones containing the C. jejuni ORFs in the vector pTLJ03 are available from Open Biosystems. pTLJ03 is an expression vector that generates N-terminal GST-His-tagged fusion proteins from the C. jejuni ORF set. Plasmid maps, details, and sequences are either available below or at http://proteome.wayne.edu. Recombination sites were verified by sequencing, and most clones have been 5’ sequenced. The ORFs range from 45 – 4,020bp in length.

Note: pTLJ03 is not a high-copy number plasmid. The C. jejuni ORF clones are cultured in LB with 100ug/ml Ampicillin.

E. coli Promoters
an Open Labs Product for investigating gene regulation

Researchers at the Weizmann Institute of Science have produced a collection of E. coli strains that enables monitoring of gene expression at high temporal resolution in living cells. Each of the reporter strains has a bright, fast-folding green fluorescent protein (GFP) fused to a full-length copy of an E. coli promoter in a low-copy plasmid.

This collection includes more than 1900 promoters (out of 2500 in the entire genome) for E. coli K12 strain MG1655 and enables measurement of gene expression at a resolution of minutes with high accuracy and reproducibility. Performing experiments in a multi-well fluorimeter using FACS or time-lapse fluorescence microscopy has the necessary sensitivity to measure gene expression in individual cells.
