HIV-1 BaL PURIFIED VIRAL LYSATE

 

PRODUCT DESCRIPTION:
The HIV-1 BaL virus is propagated in PM-1 cells, a clonal derivative of Hut78 which allows for the growth of macrophage and T-cell tropic viruses. The virus is purified using density gradient ultracentrifugation, disrupted in the presence of detergent and characterized at the biochemical and immunologic levels to assure product consistency and performance.

 

PRODUCT COMPOSITION:
Purified HIV-1 (density 1.16g/cm3) is disrupted in 0.5% Triton X-100 nonionic detergent/0.6M KCI. Detergent is removed by 2 cycles of ether extraction and the virus is subjected to in vitro testing to ensure lack of infectivity. Each lot of purified virus is examined for purity using SDS-PAGE and for antigen composition using Western immunoblotting (WIB). Diagnostic levels of env proteins (gp160, gp120 and gp41), gag proteins (p17, p24, p39, p55) and pol gene-derived proteins (p31, p51/p66) are present in the virus. The total protein concentration is determined by Pierce BCA assay. Verification of virus inactivation is determined by Reverse Transcriptase Reduction Assay.

 

RECOMMENDED USES:
Immunodetection of antibodies to HIV-1 using solid-phase enzyme immunoassay (EIA), WIB, dot-blotting and related techniques. The material should also be of value as an immunogen and for the purification of viral proteins.

 

STORAGE AND STABILITY:
The purified virus is stable for at least one year when stored at -70°C. To avoid freeze-thawing, unused material should be stored in aliquots, preferably under nitrogen in order to prevent oxidation.

 

AVAILABILITY:
Distributed in vials containing 1.0 mg of viral protein, shipped on dry ice. Protein concentration ranges from 0.5 to 1.5 mg/ml. Custom orders are available, including (1) specific buffer formulations, (2) package sizes, (3) solid-phase adsorption to agarose, plastic, etc. and (4) conjugation to fluorochromes (FITC, rhodamine, etc.), enzymes, biotin, streptavidin, etc.
