INTENDED
USE |
|
Glutathione (GSH) acts as an antioxidant and is oxidized to GSSG in
biological systems. The Stabilization Solution I is provided to
stabilize reduced glutathione (GSH) in samples.
* The Stabilizing Solution I is for Research Purposes Only.
The reagent provided is enough to stabilize ~ 50 aliquots (0.1 ml)
of EDTA whole blood.

PRECAUTIONS |
|
· Please
read all instructions carefully.
· To avoid cross contamination, use separate pipette tips for each
specimen.
· Universal safety precautions while working with bio-hazardous
materials should be adopted (1).
· Wear gloves, lab coats and safety glasses at all times.
· All contaminated materials should be properly disposed and work
surfaces appropriately decontaminated. |

Blood Draw
Requirements:
The following evacuated blood tubes should be drawn by standard
venipuncture procedure:
1-5 mL Lavender Top, Liquid K3 EDTA, 15% additive
solution.
Care should be taken that blood tubes are completely filled.
Invert blood tubes a minimum of five times for proper distribution
of anticoagulant. |

MATERIALS |
|
Pipetters
Centrifuge (capable of 1500 X g)
1.8 mL Cryovials with sealing gasket or 2 mL snap cap micro
centrifuge tubes.
Freezer at –20 ºC or lower
Stabilization Solution I (Catalog #0805003) |

METHODS |
|
Glutathione Stabilization
1) Add 0.1 mL EDTA whole blood to 1.9 mL Stabilization Solution I.
2) Vortex and leave at RT for 5 minutes.
3) Centrifuge @ 1500 X g for 10 minutes.
4) Decant supernatant to 1.8 mL cryovial
5) Freeze ASAP at –80 ºC or lower.
Label samples with ID#, specimen type and date of preparation.
Note: The time between phlebotomy and freezing of specimen should
not exceed 90 min.
Samples are stable for at least 12 months when stored at –80º C with
no thawing.
Note: An extra sample of chilled EDTA whole blood or a spun
hematocrit value has to be provided for normalization. |
