Selective Profiling of Kinase Inhibitors
The PepChip array facilitates drug discovery by identifying
kinase inhibitor specificity. With the PepChip array 1,152
peptide kinase substrates can be screened in one experiment.
Multiplex Kinase Profiling with the PepChip® Kinase Array
A comparison of kinase patterns between normal and disease
state or untreated vs. treated cells can be obtained by using
the PepChip Kinase Array. It is also a useful tool to
investigate the specificity of a kinase inhibitor in the context
of the total kinase activity of the target cells using cell
lysates.
Substrate Determination with PepChip® Kinase
Substrates for novel and known kinases are often determined
using kinase assays. Traditionally, kinase assays are performed
with only a few potential peptide or protein substrates. With
PepChip Kinase you can screen 1,152 unique substrates in one
experiment.
Synthetic Peptides Ensure Array-to-array Consistency
The PepChip Kinase array was designed for drug development
and signal transduction research. The array consists of 1,152
synthetic kinase substrate peptides covalently bound to the
surface of a treated glass slide. Synthetic peptides are highly
stable and very consistent in structure, eliminating the
problems that result when traditional protein array content is
used.

One million serum-starved cultured cells were left untreated
(left panel), incubated with growth factor (middle panel) or
incubated with growth factor and a specific small molecule
tyrosine kinase inhibitor (right panel). After a 10 minute
incubation, lysates were prepared and cellular kinome patterns
were determined using PepChip Kinase microarrays. Statistical
analysis showed that phosphorylation of 7% of the substrates
changed significantly as a result of the treatment.
Layout of the PepChip Kinase Slides
The PepChip Kinase Slide contains peptides arrayed on a 25 x
75 mm glass slide. The array design contains two identical
arrays of 1,176 peptides, including one rhodamine-labeled
peptide per subarray used for quality control purposes, arrayed
in a pattern of 4 x 6 subarrays. The PepChip Kinase Evaluation
Slide contains two identical arrays of 192 peptides in a pattern
of 2 x 4 subarrays containing 24 peptides. Test slides are
available for optimization of reaction conditions.
