CometAssay™ provides reagents
and our exclusive CometSlide™
for the rapid analysis of DNA fragmentation associated with DNA
damage. The comet assay or single cell gel electrophoresis assay
is based on the alkaline lysis of labile DNA at sites of damage.
The unwound, relaxed DNA is able to migrate out of the cell
during electrophoresis and can be visualized using **SYBR®
Green I nucleic acid gel stain. Cells that have accumulated DNA
damage appear as fluorescent comets with tails of DNA
fragmentation or unwinding, whereas, normal undamaged DNA does
not migrate far from the origin. The CometAssay is provided with
our exclusive CometSlides which greatly simplify the assay. Each
slide provides two sample surfaces specially treated to promote
agarose adherence, and a hydrophobic barrier to allow treatment
with one of Trevigen’s DNA repair enzymes. Simply add your cells
to the low melting point Comet LMAgarose, and pipet onto the
slide. |

agarose. This eliminates the time consuming and unreliable
traditional method of preparing base layers of agarose. The use
of CometSlide 96 shortens assay time, and allows the rapid and
reliable analysis of large numbers of samples. Trevigen also
provides **SYBR® Green I for
DNA visualization providing improved sensitivity compared to
ethidium bromide. The CometAssay may be coupled with Trevigen’s
FLARE™ (Fragment Length
Analysis using Repair Enzymes) Assay that provides the added
ability to probe for specific types of DNA damage using DNA

Trevigen’s
CometAssay™ Silver Staining
Kit is designed for the convenient silver staining of CometAssay™
or single cell gel electrophoresis results. Recording of data
generated by the CometAssay™
and staining with fluorescent dyes requires epifluorescence
microscopy and rapid analysis because of the generally short
lifespan of the dye. Using the Silver Staining kit, permanent
records are created and visualization using standard light
microscopy is possible. The Silver Staining Kit was designed
specifically for use with the CometSlides™
to minimize unwanted background and the amount of hazardous
waste generated by silver nitrate.

| Trevigen's new
comet assay kit, DASH™
(Diffusion Apoptosis Slide Halo), allows discrimination of
healthy and damaged cells in just a few hours. Simply embed
cells in low melting agarose on a pre-treated slide, lyse under
alkaline conditions, and precipitate the DNA of the remaining
nuclear region (i.e. nucleoid) in the agarose. The precipitated
DNA is visualized using **SYBR®
Green I nucleic acid stain. When excited (425 - 500 nm) the
DNA-bound **SYBR® Green I
emits green light. In healthy cells, the fluorescence is
confined to the nucleoid: undamaged DNA is supercoiled and thus
does not diffuse very far, if at all, from the nucleoid. In
cells that have accrued damage to the DNA, the alkali lysis
treatment unwinds the DNA, releasing fragments that diffuse away
from the center of the nucleoid, creating a "halo" surrounding a
small compact origin. This kit includes ready to use, pre-coated
slides and optimized reagents for your convenience. |
