Anti-human/ mouse Caspase 2 Polyclonal Antibody, 40 µg |
Following
induction of apoptosis, proteolytic cleavage of procaspase-3
occurs to generate an active 18 kDa caspase-3 fragment which
targets key modulators of the apoptotic pathway, including
poly-ADP-ribose polymerase and other caspases, for cleavage.
Trevigen’s antibody recognizes the active 18 kDa fragment
but does not recognize the 32 kDa procaspase-3, and is
therefore useful as a specific marker for apoptosis.

acid peptide sequence from carboxyl terminus of the human

Caspase 9 is
an effector caspase involved in the activation of caspases
including caspase 3. The procaspase 9 binds ATP and
cytochrome c, and subsequently proteolytically cleaves
itself to form the active caspase 9. Caspase 9 is thought to
be involved in mitochondrial regulation of apoptosis.

| Apoptosis
can be induced by the engagement of the death receptor CD95
(APO-1/Fas). Signaling through Fas requires Fas aggregation
and the formation of an intracellular protein complex called
the death-inducing signaling complex. This complex consists
of the cytoplasmic domain of Fas, the adaptor molecule
FADD/MORT-1 and procaspase-8 (Mch5/Flice/Mach). |

APAF-1
(apoptotic protease activating factor-1) is a mammalian
homolog of CED-4 that regulates cell death by participating
in a ternary complex with cytochrome c, and procas-pase-9.
APAF-1 is a member of the CARD protein family, which
contains a caspase-recruitment domain linked to a
nucleotide-binding domain (NBD), regulating apoptosis and/or
NF-ê B activation. During apoptosis, release of cytochrome c
initiates dATP-dependent oligomerization of APAF-1 and
formation of the apoptosome caspase processing complex,
triggering the postmitochondrial-mediated effector caspase
cascade.

The
abundance of glyceraldehyde-3-phosphate dehydrogenase
(G3PDH) in eukaryotic cells is relatively unaffected by
external factors. In Western blot analysis the level of
G3PDH can be used as a reference value for comparison
between different cell lysates or gel loadings.
|

rabbit polyclonal antibody raised against a synthetic
peptide corresponding to a portion of the human
glyceraldehyde-3-phosphate dehydrogenase gene. |

| Members of
the IAP (Inhibitor of Apoptosis Protein) family were
initially identified in baculoviruses. IAPs are found in a
wide variety of organisms from vertebrates to insects
(Drosophila), and yeast to invertebrates (Caenorhabditis
elegans). XIAP, cIAP-1, and cIAP-2 are thought to inhibit
apoptosis through direct caspase inhibition. The
anti-apoptotic range of action of IAPs is broad and overlaps
with Bcl-2 family activity. All IAP proteins contain at
least one 70 amino acid baculovirus IAP repeats (BIR) which
is essential for their anti-cell death activity that is
thought to work by protein-protein interaction via zinc
binding domain. A common characteristic of the
caspase-binding IAP proteins (XIAP, cIAP-1 and cIAP-2) is
that they all exhibit three BIR domains in their sequence.
|
| |
| cIAP-1 is the mammalian IAP
homolog B (MIHB). This protein contains three tandem BIR
domains and is believed to bind specifically to caspase 3
and caspase 7 and not to the early apoptosis effector
caspases 8, 1, or 6. |
| |
| Anti-cIAP-1 is a
rabbit polyclonal antibody generated by immunization with a
KLH-coupled synthetic peptide corresponding to amino acids
527 to 546 of the human cIAP-1 sequence. |
| |
| The antibody
recognizes human and mouse cIAP-1, and will not recognize
cIAP-2 on Western blots. |

TRAIL (Tumor
Necrosis Factor-Related Apoptosis Inducing Ligand) is a cell
surface molecule involved in the initiation of apoptosis.
Known as a death receptor like Fas, TNFR-2, and DR-3, TRAIL
is activated upon ligand binding and engages the cells on
the cascade of signaling involving caspase activation
through an adapter molecule. TRAIL has been shown to induce
apoptosis in human melanoma cells and not in healthy cells
making it a prime candidate for cancer studies.

goat polyclonal antibody generated by immunization with
full-length recombinant human TRAIL. |

recognizes and neutralizes human TRAIL. |

specifically to caspase 3 and caspase 7. |
