Bax, a 21 kDa eukaryotic protein plays an important role in the regulation of cell death in a number of eukaryotic cells. The over-expression of Bax has been shown to accelerate cell death. The ratio of Bax to other Bcl-2 family members, and its subcellular distribution is thought to help regulate the process of programmed cell death.

Bax is a 24 kDa protein that shares the BH1, BH2, BH3 domain of Bax, but has a unique carboxyl terminus that lacks the hydrophobic transmembrane domain. The distribution of Bax mRNA differs from that of Bax RNA, and is particularly abundant in the brain.
   
Immunogen: The anti-Bax is a sheep polyclonal antibody raised against a synthetic peptide corresponding to amino acids 172 to 199 of the Bax protein.
   
Specificity: The antibody recognizes human Baxß.
   
Preparation: The antibody is affinity purified and provided in phosphate buffered saline containing less than 0.1% sodium azide.

The Bcl-2 family of proteins plays a crucial role in the regulation of cell death in many eukaryotic systems. Bcl-XL is a 28 kDa protein associated with cell survival. Bcl-X is reported in two forms, Bcl-XL and Bcl-XS. The short form is reported to inhibit the survival promoting activity of the long form. Bcl-XS is a splice variant at the RNA level, but its functional role is not clear.
   
Immunogen: The antibody was raised against a synthetic peptide corresponding to amino acids 3 to 14 of the human Bcl-XL sequence.
   
Specificity: The YTH-2H12 clone recognizes human, mouse and rat Bcl-XL protein.

Bim (Bcl-2 interacting mediator of cell death) is a proapoptotic protein sharing only the short BH3 domain (Bcl-2 Homology regions) with other members of the Bcl-2 family. Bim has three known isoforms, thought to be generated by alternative splicing. Bim is essential for certain apoptotic responses and to prevent overproduction of hematopoietic cells suggesting that it may be a tumor suppressor.

Cytochrome c is a well-characterized mitochondrial protein involved in cellular energy metabolism. When released into the cytosol, cytochrome c can activate the caspases responsible for apoptosis through interaction with protease activating factors.

The precursor of cytochrome c, apocytochrome c, is synthesized in the cytoplasm. Upon translocation into the mitochondria, cytochrome c refolds and acquires a heme moiety required for functionality in the mitochondrial respiration chain. The heme-bound form of cytochrome c is called holocytochrome c.

Mcl-1 is a Bcl-2 homolog involved in early induction of cellular differentiation. Cloned from the human myeloblastic leukemia cell line, Mcl-1 is a 40 kDa mitochondrial protein that has been shown to inhibit apoptosis through the formation of dimers with Bcl-2 family member Bax. Mcl-1 is expressed in a wide variety of cancerous tissues.
 
Immunogen: Anti-Mcl-1 is a rabbit polyclonal antibody generated by immunization with a KLH-coupled synthetic peptide corresponding to amino acids 175 to 194 of the human Mcl-1 sequence. The murine sequence differs by a single amino acid substitution of S for A at position 192 of human Mcl-1.
   
Specificity: The antibody recognizes human Mcl-1. The antibody also detects a 27-34 kDa band in extracts of mouse L929 and DA3 cells believed to be the murine form of Mcl-1.
   
Preparation: The antibody is affinity purified and provided in sterile PBS (pH 7.4) containing 0.01% sodium azide.

The peripheral-type benzodiazepine receptor (PBR) is a ubiquitous protein involved in the regulation of cholesterol transport from the outer to inner mitochondrial membrane. Thought to play a role in the formation of the mitochondrial permeability transition (PT) pore involved in apoptotic mechanisms, PBR is also present in abundance in tumors, in aggressive breast cancer cell lines, and in metastatic human breast tumor biopsy sections.
Immunogen: The anti-PBR antibody was developed in rabbits against an internal sequence of the PBR protein consisting of amino acids 71 to 88 of the human PBR sequence. The specificity of the antibody was assessed in steroidogenic cells and in mitochondrial extracts.
   
Specificity: The antibody recognizes mouse, rat, and human PBR protein.
   
Preparation: Anti-PBR antibody is provided as rabbit anti-seral.
   
Applications: • Immunohistochemistry
• Western blotting
• Starting recommended dilution for Western blot is 1:1,000; for immunohistochemistry, 1:250.
