Description: Complex of N-terminal GST-tagged recombinant full-length human p110alpha ( GenBank Accession No.U79143), and N-terminal His tagged, recombinant full length, human p85alpha (XM_043865). Coexpressed in a Baculovirus infected Sf9 cell expression system. p110alpha MW=155 kDa, p85alpha MW=88 kDa.

Formulated in: 25 mM Tris-HCl, pH 8.0, 100 mM NaCl, 0.05% Tween-20, 50% glycerol, and 3 mM DTT.

Stability: >6 months at –80°C

Purity: >80% by SDS-PAGE .

Enzyme Activity: p110alpha/p85alpha activity was determined using PIP2 and ATP as substrates. Assay buffer: 50 mM HEPES, pH 7.4, 150 mM NaCl, 5 mM MgCl2, 1 mM DTT, 3 uM Na-orthovanadate, 0.1 mM ATP, 1 uM PIP2, and 6 ng/ul p110alpha/p85alpha. The enzyme at concentration 6 ng/ul converts about 30% of 1 uM PIP2 to PIP3 at 22°C for 60 min.

Application: useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
