Membrane-permeant form and thus can be loaded into cells via incubation. Because of the relatively low water solubility of the AM ester, Pluronic F-127 (See 59000 and refs. Therein), a mild detergent, is often used as a dispersing agent to facilitate the loading. Rhod-2/AM itself does not bind Ca²⁺, but it is readily hydrolyzed to rhod-2 by endogenous esterases once the dye is inside the cells.

EDC (or EDAC) has recently been found to be very useful for fixing in situ chelators, including the fluorescent ion indicators in this catalogue. The fixation of ion indicators makes it useful for post histological studies following the physiological experiments. EDC is also a widely used reagent to activate carboxy groups for amine couplings.
