Description:
Apoptosis and necrosis are two major
processes by which cells die. Apoptosis is an active,
genetically regulated disassembly of the cell from within.
Disassembly creates changes in the phospholipid content of

(PS) is translocated from the inner to the outer surface of
the cell for phagocytic cell recognition. The human

Caspase-3 is a cell-death protease
involved in the execution phase of apoptosis (1,2).
Caspase-3 Luminescent Assay Kit provides a homogenous assay
system for fast and ultra-sensitive detection of caspase-3
activity in purified enzyme or mammalian cell systems. This
kit utilizes Ac- DEVD-amino-D-luciferin in combination with
Firefly luciferase to detect caspase-3 activity. Upon
substrate cleavage at the Cterminal side of the DEVD peptide
sequence by caspase-3, amino-D-luciferin is released.
Amino-D-luciferin is a synthetic derivative of the natural
luciferase substrate D-luciferin and is also accepted by the
enzyme as a substrate, capable of generating bioluminescence
in the presence of oxygen, ATP and Mg2+. Thus,
amino-D-luciferin released from the caspase-3 activity is
subsequently oxidized via Firefly luciferase activity with
emission of light, the intensity of which is proportional to
the caspase-3 activity.
The history of detecting peptidase activities via
bioluminescence goes back a decade ago when Monsees and
colleagues first demonstrated the principle of coupling
peptidase activity to Firefly luciferase activity by the use
of amino-D-luciferin (3,4). The authors showed
that virtually any bioluminogenic peptidase substrates could
be made by conjugating a known peptidase-recognition amino
acid or peptide sequence to the amine group of
amino-D-luciferin, a method similar to how the widely used
chromogenic and fluorogenic peptidase substrates are
constructed. Therefore, considering the superior sensitivity
of bioluminescence-based detections, we have developed a
line of caspase and other peptidase substrates using the
same peptide sequences used in our chromogenic and
fluorogenic caspase substrates (See Assay Principle below).
Under optimal conditions, it is possible to detect <1 ng of
a peptidase by using a bioluminogenic substrate.

Description:
Caspase-9 LEHD-R110 Fluorometric HTS
Assay Kit is specifically designed for HTS-based assays. The
kit provides a homogenous assay system for fast and highly
sensitive detection of caspase-9 activity by fluorescence in
mammalian cells. The fluorogenic substrate (Ac-LEHD)2-R110
contains two LEHD tetrapeptides and is completely hydrolyzed
by the enzyme in two successive steps. Cleavage of the first
LEHD peptide results in the monopeptide Ac-LEHDR110
intermediate, which has absorption and emission wavelengths
similar to those of R110 (labs/lem
= 496/520 nm) but has only about 10% of the fluorescence of
the latter. Hydrolysis of the second LEHD peptide releases
the dye R110, leading to a substantial fluorescence increase
(See Figure B3 for assay principle).
The assay kit includes Ac-LEHD-CHO, which is a caspase-9
inhibitor and can be used as a negative control. Also, R110
is provided in the kit for generating a standard curve,
which can be used for quantifying caspase-9 activity.

Description:
The loss of mitochondrial membrane
potential is a hallmark for apoptosis. It is an early event
preceding phosphatidylserine externalization and coinciding
with caspase activation 1-2. JC-1 Mitochondrial
Membrane Potential Detection Kit is used to measure
mitochondrial membrane potential changes in cells. In
non-apoptotic cells, JC-1 accumulates as aggregates in the
mitochondrial membranes, resulting in red fluorescence (590
nm). The brightness of red fluorescence is proportional to
the potential and varies among different cell types.
However, in apoptotic and necrotic cells, which have
diminished mitochondrial membrane potential,JC-1 exists in
the green fluorescent (529 nm) monomeric form 3-5.
This kit provides a step-by-step protocol and ready-to-use
reagents for performing >100 assays for use on flow
cytometers, fluorescence microscopes and fluorometric plate
readers.

Description:
Diminished cellular glutathione (GSH)
level occurs at the early stage of mitochondrion-associated
apoptosis pathway due to GSH efflux. GSH depletion further
leads to cytochrome c release and caspase 3 induction (1,2)
MCB Glutathione Detection Kit utilizes a thiol-reactive dye
monochlorobimane (MCB), which is essentially monfluorescent
until it reacts with a thiol to form a blue fluorescent
product (labs/lem
= 380/461 nm). By incubating cellular lysate with MCB, the
intensity of the fluorescent signal generated from the assay
reflects the amount of GSH present in the cells.
Fluorescence can easily be detected using a fluorometer or
96- well fluorometric plate reader.
