Apoptosis Assays

Catelog Number :
    30017
Product Name :
    Apoptosis & Necrosis Quantification Kit

Kit Components:

  • FITC-Annexin V
  • Ethidium homodimer III
  • 5X binding buffer

Description:

Apoptosis and necrosis are two major processes by which cells die. Apoptosis is an active, genetically regulated disassembly of the cell from within. Disassembly creates changes in the phospholipid content of the cytoplasmic membrane outer leaflet. Phosphatidylserine (PS) is translocated from the inner to the outer surface of the cell for phagocytic cell recognition. The human anticoagulant, annexin V, is a 35 kD Ca2+-dependent phospholipid protein with a high affinity for PS. Annexin V labeled with fluorescein (FITC) (labs/lem= 492/ 514 nm) can identify apoptotic cells in green by binding to PS exposed on the outer leaflet. Necrosis normally results from a severe cellular insult. Both internal organelle and plasma membrane integrity are lost, resulting in spilling of cytosolic and organellar contents into the surrounding environment. Ethidium homodimer III (EtD-III) is a highly positivelycharged nucleic acid probe, which is impermeant to live cells or apoptotic cells, but stains necrotic cells with red fluorescence (labs/lem(with DNA) = 528/617 nm). EthD-III is a superior alternative to propidium iodide (PI) or ethidium homodimer I used in some of our competitors?kits because of its significantly higher affinity for DNA and higher fluorescence quantum yield. Apoptosis & Necrosis Quantification Kit provides a convenient assay for quantifying apoptotic (green) and necrotic (red) cells within the same cell population by flow cytometry or fluorescence microscopy.

Features:

  • Simple: Simultaneous detections of both apoptotic and necrotic cells in a single assay.
  • Fast: A single 15-30 min staining step.
  • Sensitive: EthD-III is a superior alternative to both PI and EthD-I for staining necrotic cells.

Spectra

For spectra of fluorescein and EthD-III.

Product Name :
    Caspase-3 Bioluminescent Assay Kit

Kit Components:

  • Cell lysis/assay buffer
  • Enzyme substrate Ac-DEVD-amino-D-luciferin
  • Enzyme inhibitor Ac-DEVD-CHO

Description:

Caspase-3 is a cell-death protease involved in the execution phase of apoptosis (1,2). Caspase-3 Luminescent Assay Kit provides a homogenous assay system for fast and ultra-sensitive detection of caspase-3 activity in purified enzyme or mammalian cell systems. This kit utilizes Ac- DEVD-amino-D-luciferin in combination with Firefly luciferase to detect caspase-3 activity. Upon substrate cleavage at the Cterminal side of the DEVD peptide sequence by caspase-3, amino-D-luciferin is released. Amino-D-luciferin is a synthetic derivative of the natural luciferase substrate D-luciferin and is also accepted by the enzyme as a substrate, capable of generating bioluminescence in the presence of oxygen, ATP and Mg2+. Thus, amino-D-luciferin released from the caspase-3 activity is subsequently oxidized via Firefly luciferase activity with emission of light, the intensity of which is proportional to the caspase-3 activity.

The history of detecting peptidase activities via bioluminescence goes back a decade ago when Monsees and colleagues first demonstrated the principle of coupling peptidase activity to Firefly luciferase activity by the use of amino-D-luciferin (3,4). The authors showed that virtually any bioluminogenic peptidase substrates could be made by conjugating a known peptidase-recognition amino acid or peptide sequence to the amine group of amino-D-luciferin, a method similar to how the widely used chromogenic and fluorogenic peptidase substrates are constructed. Therefore, considering the superior sensitivity of bioluminescence-based detections, we have developed a line of caspase and other peptidase substrates using the same peptide sequences used in our chromogenic and fluorogenic caspase substrates (See Assay Principle below). Under optimal conditions, it is possible to detect <1 ng of a peptidase by using a bioluminogenic substrate.

Features:

  • Ultra-sensitive: Detectable with fewer than 100 cells with excellent signal-to-noise ratio.
  • Simple: Single-step homogenous assay.
  • HTS-compatible: The extended-glow signal is compatible with HTS format.

Kit Components:

  • Cell lysis buffer
  • Assay buffer
  • Enzyme substrate (Ac-DEVD)2-R110
  • Enzyme inhibitor Ac-DEVD-CHO
  • R110

    Description:

Caspase-3 is an active cell-death protease involved in the execution phase of apoptosis, during which cells undergo morphological changes such as DNA fragmentation, chromatin condensation, and apoptotic body formation (1, 2). Caspase-3 DEVD-R110 Fluorometric and Colorimetric Assay Kit provides a simple assay system for fast and highly sensitive detection of caspase-3 activity either by fluorescence or absorbance in mammalian cells. The fluorogenic and chromogenic substrate (Ac-DEVD)2-R110 contains two DEVD tetrapeptides and is completely hydrolyzed by the enzyme in two successive steps (3, 4). Cleavage of the first DEVD peptide results in the monopeptide Ac-DEVD-R110 intermediate, which has absorption and emission wavelengths similar to those of R110 (lex\lem= 496/520 nm) but has only about 10% of the fluorescence of the latter. Hydrolysis of the second DEVD peptide releases the dye R110, leading to a substantial fluorescence increase (See Assay Principle below).

Although fluorometric detection of the end products is preferred because of the superior sensitivity, detection by absorbance is also possible. In fact, the extinction coefficient of R110 is 10 times higher than that of p-nitroaniline (pNA), a dye commonly used in chromogenic substrates, making R110- based substrates significantly more sensitive than pNA-based substrates, even by colorimetric detection.

The assay kit includes Ac-DEVD-CHO, which is a caspase-3 inhibitor and can be used as a negative control. Also, R110 is provided in the kit for generating a standard curve, which can be used for quantifying caspase-3 activity.

    Features:

    • Fast: Fast enzyme kinetics.
    • Sensitive: The enzymatic reaction forms intensely yellow colored and highly green fluorescent rhodamine 110 (R110) product. The long wave- lengths of R110 excitation and emission minimize cellular autofluorescence.
    • Versatile: Compatible with both fluorometric and colorimetric detection systems.

    • Cell lysis/assay buffer
    • Enzyme substrate Ac-IETD-amino-D-luciferin
    • Enzyme inhibitor Ac-IETD-CHO

    Description:

Caspase-8 is the most upstream caspase in the CD95/Fas apoptotic pathway and is activated by the signaling pathways for CD95/Fas and TNF. The ultra-sensitive Caspase-8 Bioluminescent Assay Kit is based on the same principle as the Caspase-3 Bioluminescent Assay Kit is based on (See description for #30010-1 and #30010-2). Kit Components:

  • Cell lysis buffer
  • Assay buffer
  • Enzyme substrate (Ac-IETD)2-R110
  • Enzyme inhibitor Ac-IETD-CHO
  • R110

    Description:

Caspase-8 is the most upstream caspase in the CD95/Fas apoptotic pathway and is activated by the signaling pathways for CD95/Fas and TNF. Caspase-8 Fluorometric and Colorimetric Assay Kit provides a simple assay system for fast and sensitive detection of caspase-8 activity either by fluorescence or absorbance in mammalian cells. The kit includes the fluorogenic substrate (Ac-IETD)2-R110, the enzyme inhibitor Ac-IETD-CHO as a negative control, R110 as a standard and optimized lysis and assay buffers. The substrate (Ac-IETD)2-R110 contains two IETD tetrapeptides and is completely hydrolyzed by the enzyme in a two-step process. Cleavage of the first IETD results in the monopeptide Ac- IETD-R110 intermediate, which has absorption and emission wavelengths similar to those of R110 (lex\lem= 496/520 nm) but has only about 10% of the fluorescence of the latter. Hydrolysis of the second IETD peptide releases the dye R110, leading to a substantial fluorescence increase (See Figure B3 for assay principle).

    Features:

    • Fast: Fast enzyme kinetics.
    • Sensitive: The enzymatic reaction forms intensely yellow colored and highly green fluorescent rhodamine 110 (R110) product. The long wave lengths of R110 excitation and emission minimize cellular autofluorescence.
    • Versatile: Compatible with both fluorometric and colorimetric detection systems.

Caspase-9 LEHD-R110 Fluorometric HTS Assay Kit is specifically designed for HTS-based assays. The kit provides a homogenous assay system for fast and highly sensitive detection of caspase-9 activity by fluorescence in mammalian cells. The fluorogenic substrate (Ac-LEHD)2-R110 contains two LEHD tetrapeptides and is completely hydrolyzed by the enzyme in two successive steps. Cleavage of the first LEHD peptide results in the monopeptide Ac-LEHDR110 intermediate, which has absorption and emission wavelengths similar to those of R110 (labs/lem = 496/520 nm) but has only about 10% of the fluorescence of the latter. Hydrolysis of the second LEHD peptide releases the dye R110, leading to a substantial fluorescence increase (See Figure B3 for assay principle).

The assay kit includes Ac-LEHD-CHO, which is a caspase-9 inhibitor and can be used as a negative control. Also, R110 is provided in the kit for generating a standard curve, which can be used for quantifying caspase-9 activity.

    Features:

HTS-compatible: Single-step homogenous assay specifically designed for HTS-based detection.
    • Fast: Fast enzyme kinetics.
    • Sensitive: The enzymatic reaction forms intensely green fluorescent rhodamine 110 (R110) product. The long wavelengths of R110 excitation and emission minimize cellular autofluorescence.

    Product Name :
JC-1 mitochondrial membrane potential detection kit

    Kit Components:

    1. 500 uL 100X JC-1

    2. 10 mL 10X buffer

    Description:

The loss of mitochondrial membrane potential is a hallmark for apoptosis. It is an early event preceding phosphatidylserine externalization and coinciding with caspase activation 1-2. JC-1 Mitochondrial Membrane Potential Detection Kit is used to measure mitochondrial membrane potential changes in cells. In non-apoptotic cells, JC-1 accumulates as aggregates in the mitochondrial membranes, resulting in red fluorescence (590 nm). The brightness of red fluorescence is proportional to the potential and varies among different cell types. However, in apoptotic and necrotic cells, which have diminished mitochondrial membrane potential,JC-1 exists in the green fluorescent (529 nm) monomeric form 3-5. This kit provides a step-by-step protocol and ready-to-use reagents for performing >100 assays for use on flow cytometers, fluorescence microscopes and fluorometric plate readers.

    Features:

    • Fast and simple: 15 minutes of incubation with the dye, followed by washing and measurement.
    • Cell membrane-permeable: Direct detection of apoptosis and mitochondrial membrane potential on live cells.
    • Versatile: Detection compatible with flow cytometers, fluorescence microscopes and fluorometric plate readers.

    Catelog Number :
        30019
    Product Name :
        MCB Glutathione Detection Kit

    Kit Components:

    • Cell lysis buffer
    • Monochlorobimane (MCB)
    • GST postive control

    Description:

Diminished cellular glutathione (GSH) level occurs at the early stage of mitochondrion-associated apoptosis pathway due to GSH efflux. GSH depletion further leads to cytochrome c release and caspase 3 induction (1,2) MCB Glutathione Detection Kit utilizes a thiol-reactive dye monochlorobimane (MCB), which is essentially monfluorescent until it reacts with a thiol to form a blue fluorescent product (labs/lem = 380/461 nm). By incubating cellular lysate with MCB, the intensity of the fluorescent signal generated from the assay reflects the amount of GSH present in the cells. Fluorescence can easily be detected using a fluorometer or 96- well fluorometric plate reader.

    Features:

    • Simple: Compatible with a standard or fluorescence plate reader.
    • Fast: Around 1 hour assay time.
