High-fidelity polymerase
developed for DHPLC usage.
DH-HiPol DNA Polymerase is a
high-performance polymerase
with high-fidelity, ideal
for mutation detection
applications. It has been
optimised for use in
detergent-free systems,
making it ideally suited for
loading samples directly
onto DHPLC columns. In any
analysis of DNA sequence
variation, it is imperative
that the polymerase used
exhibits high fidelity.
Central to the detection of
mutations by DHPLC is the
comparative analysis of
paired
homoduplex-heteroduplex
mixtures, arising at the
position of sequence
variation. The presence of
inappropriate heteroduplexes
will confound the
interpretation of
experimental data and
therefore polymerase-induced
misincorporation must be
eliminated as far as
possible. Proof-reading
enzymes have a dramatically
lower misincorporation rate
owing to the presence of a
3’-5’ "proof-reading"
exonuclease domain. In
contrast to detergent-free
polymerases from other
manufacturers, DH-HiPol is a
single proof-reading enzyme
which does not contain or
require the addition of
non-proof-reading enzyme to
enhance activity.

DH-HiPol can be stored for
12 months at -20°C in a
constant-temperature
freezer.

DH-HotPol DNA Polymerase is
a heat-activated version of
our DH-HiPol Polymerase. It
has the same proof-reading
qualities and features as
DH-HiPol, but with the added
safety and reliability
inherent in heat-activated
polymerases. It is ideal for
DHPLC applications where
mispriming can cause
problems with the
interpretation of results
and is designed for use in
high-throughput
applications, where longer
set-up times can lead to
lower specificity.

Ultimate Resolution Test
Sample.
DH-Ladder is designed for
running pre-, during- and
post-DHPLC, to determine if
the sensitivity is uniform
throughout the multi-sample
run being analysed. It has 6
peaks arranged in 3 pairs,
allowing resolution to be
determined for small, medium
and large fragments.
The peaks are 95, 105, 255,
270, 440 and 470 bp.

Before and during each run
it is essential to determine
the resolution of the
column. This procedure is
carried out under
non-denaturing conditions
(50ºC). A typical control
run would include a range of
fragments, with several DNA
products of a similar size.
The DNA fragments are: 11,
18, 80, 102, 174, 257, 267,
298, 434, 458 and 587 base
pairs.
