* HAT - /HT - Supplement
 
  • Hybridoma selection using HAT/HT

    Basal media suitable for lymphoid cells (e.g. RPMI 1640, Dulbecco's MEM), supplemented with (50x) HAT/HT concentrate, can serve as selection media for hybridoma cells.

    Metabolic precursors, 100 µM Hypoxanthine (for purine synthesis), 16 µM Thymidine (for pyrimidine synthesis), both able to serve the side-pathway, and 0.4 µM Aminopterine (as a folic acid antagonist, inhibitor of the main pathway of nucleic acid synthesis) are added to the medium.

    By fusing normal lymphocytes with transformed, gene-defect myeloma cells - e.g. missing hypoxanthine-guanine-phosphoribosyl-transferase activity (HGPRT-) and/or missing thymidine kinase activity (TK- - only hybridomas of both cells can survive, and proliferate.

    B-lymphocytes from the spleen of an immunized animal, will survive only for a few days and do not proliferate, although by their intact metabolic side pathway (HGPRT+, HT+ and the addition of hypoxanthine and thymidine, they could bypass the blockade by aminopterin.

    Cells with retained proliferate potential will eventually die off because of their enzyme deficiency (HGPRT-, T>K-.

    Only hybridomas 'repaired' by genetic complementation (HGPRT+, TK+ through fusion, will proliferate by the double selection mechanism like transformed myeloma cells.

    Successive change of the medium containing (50x) HAT, and replacing it by HT concentrate, will revert the aminopterine blockade.
