The Assay Designs' Phospholipase A2 Kit is designed to allow
the rapid detection of phospholipase A2 activity in a variety
of samples. Secretory Phospholipase A2's (sPLA2)
are a group of related enzymes that, upon activation, cleave an acyl
ester bond in the Sn-2 position in glycerophospholipid. These are
extracellular enzymes that have high disulfide bond content, low
molecular mass (14KDa), and require mM levels of Ca2+ for
catalysis. More importantly, these enzymes play a crucial role in the
generation of arachidonic acid. PLA2 has been shown to have a
number of biological actions including immunological responses,
inflammation, cellular proliferation, vasoconstriction, and
bronchioconstriction.

1. Pipet Standards and Samples in duplicate into the wells.
2. Pipet Reaction Buffer and Substrate into the wells and incubate.
3. Pipet Stop Reagent followed by Color Reagent into the wells and
incubate.
4. Read on a plate reader at 405 nm.
5. Calculate sample concentrations from Standard Curve.
