Smac (Second mitochondria-derived activator of caspase) is also know as
DIABLO (Direct IAP-Binding Protein with Low pI) and is commonly referred
to as Smac/DIABLO. Smac/DIABLO promotes Caspase-9 activation by binding
to inhibitor of apoptosis proteins (IAPs) and removing their inhibitory
activity. Mitochondrial import and cleavage of its signal peptide are
required for Smac/DIABLO to gain its apoptotic activity. Smac/DIABLO, a
239 amino acid protein, exits the mitochondria and enters the cytosol
during certain apoptosis events. Overexpression of Smac/DIABLO increases
the sensitivity of the cell to apoptotic stimuli. Changes in expression
levels of Smac/DIABLO have been found in a number of human cancers
including melanoma and cancers of the ovary and prostate.
1. Pipet Standards and Samples in duplicate into the wells and incubate.
2. Aspirate and wash the plate.
3. Pipet the yellow Antibody into the wells and incubate.
4. Aspirate and wash the plate.
5. Pipet the blue Conjugate into the wells and incubtate.
6. Aspirate and wash the plate.
7. Pipet Substrate into the wells and incubate.
8. Pipet Stop solution and read plate on a plate reader at 450nm.
9. Calculate sample concentrations from the Standard Curve.
