Protein G, a cell wall protein originally isolated from Type G
Streptococci, is a monomeric protein with a molecular weight of about
50,000 kDa, completely lacking in cysteine residues. Protein G binds the
Fc region of immunoglobulin G (IgG) with high affinity. Due to this
binding ability, Protein G can be used to facilitate the purification
and recovery of either polyclonal or monoclonal immunoglobulins. Protein
G will bind immunoglobulin G from many different species including all
human and mouse subclasses, binding the Fc regions of two IgG molecules
simultaneously. Protein G binds IgG from rat but substantially weaker
than with other species, and will not bind to the Fc portion of chicken
antibodies at all.

1. Pipet Standards and Samples in duplicate into the wells.
2. Pipet Antibody into the wells, then incubate.
3. Aspirate and wash the plate.
4. Pipet substrate into the wells and incubate.
5. Pipet Stop Solution and read on a plate reader at 450nm.
6. Calculate sample concentrations from Standard Curve.
