p27 is also known as, KIP1, p27Kip1, cyclin-dependent kinase
inhibitor 1B (CDKN1B), and cyclin-dependent kinase inhibitor p27. It is
composed of 198 amino acids and has a calculated molecular weight of 22
kDa. p27 is a member of a family of CDK inhibitors (CDIs) that bind to
cyclin/CDK complexes and arrest cell division. In general, p27
expression is highest in quiescent cells and falls as cells reenter the
cell cycle. p27 was first identified as an inhibitor of cyclin E-CDK2.
Several anti-proliferative signals result in the accumulation of p27.
These include cell to cell contact, the withdrawal of cytokines, the
withdrawal of mitogens and exposure to cAMP. p27 has been implicated in
several fundamental cellular processes including apoptosis, cell
division and proliferation. p27 is a tumor-suppressor gene that plays a
crucial role in the pathogenesis of human malignancy. A decrease in p27
expression has been shown to be associated with aggressive growth of
breast, lung, colorectal, gastric and prostate tumors.

1. Pipet Standards and Samples in duplicate into the wells and incubate.
2. Aspirate and wash the plate.
3. Pipet the yellow Antibody into the wells and incubate.
4. Aspirate and wash the plate.
5. Pipet the blue Conjugate into the wells and incubtate.
6. Aspirate and wash the plate.
7. Pipet Substrate into the wells and incubate.
8. Pipet Stop solution and read plate on a plate reader at 450nm.
9. Calculate sample concentrations from the Standard Curve.
