Assay Designs´ Direct cyclic GMP (cGMP) kit is designed for use with
tissue samples that have been grown in tissue culture media. The kit
includes 0.1M HCl which is added to cells to lyse them, stop endogenous
phosphodiesterase activity and stabilize the released cyclic GMP. The
0.1M HCl treated samples are then analyzed directly in the microtiter
plate without extraction, drying or reconstitution. If analysis is to be
performed on samples in buffer or with samples that have been extracted
and dried down, please refer to Assay Designs´ Regular cyclic GMP kit.
Guanosine 3´, 5´-cyclic monophosphate (cyclic GMP; cGMP) was
identified in 1963. It has been shown to be present at levels typically
10 to 100 fold lower than
cyclic AMP in most tissues and is formed by the action of the enzyme
guanylate cyclase on GTP. It is involved in a number of important
biological reactions. Some hormones, such as acetylcholine, insulin and
oxytocin, as well as certain other chemicals like serotonin and
histamine cause an increase in cyclic GMP levels. Stimulators of
guanylate cyclase such as the vasodilators nitroprusside, nitroglycerin,
sodium nitrate and
nitric oxide (NO) also stimulate cyclic GMP levels. Peptides such as
atrial natriuretic peptide (ANP) that relax smooth muscle also increase
cyclic GMP concentrations. cGMP has been confirmed as a second messenger
for ANP.

1. Pipet Standards and Samples in duplicate into the wells.
2. Pipet the blue Alkaline Phosphatase Conjugate followed by the yellow
Antibody into the wells and incubate.
3. Aspirate and wash the plate.
4. Pipet Substrate into the wells and incubate.
5. Pipet Stop Solution and read on a plate reader at 405 nm.
6. Calculate sample concentrations from Standard Curve.
