The human breast cancer type 1 susceptibility (BRCA-1) gene encodes a
nuclear phosphoprotein of 1863 amino acids (MW= 220 kDa). This gene
received considerable attention following the discovery that it is
mutated in ~45-50% of hereditary breast cancers. Additionally, BRCA-1
has been implicated in cases of sporadic breast cancers. BRCA-1 is
ubiquitously expressed in adult tissues, participating in many critical
cellular processes including DNA repair, cell cycle progression,
transcriptional regulation, chromatin remodeling and ubiquitination. All
of these processes are essential to maintenance of proper cellular
function and contribute to tumorigenesis when deregulated; thus, BRCA-1
has been proposed to function as a cellular caretaker.
1. Pipet Standards and Samples in duplicate into the wells and incubate.
2. Aspirate and wash the plate.
3. Pipet the yellow Antibody into the wells and incubate.
4. Aspirate and wash the plate.
5. Pipet the blue Conjugate into the wells and incubtate.
6. Aspirate and wash the plate.
7. Pipet Substrate into the wells and incubate.
8. Pipet Stop solution and read plate on a plate reader at 450nm.
9. Calculate sample concentrations from the Standard Curve.
