Endothelins (ET) are isopeptides produced by vascular endothelium that
have potent vasoconstriction activity. The peptides are encoded by three
separate genes and processed to yield 39-residue "Big ET" molecules
which are further processed to the 21 amino acid sequences designated
ET-1, ET-2 and
ET-3. Vascular endothelial injury stimulates the release of
endothelins, which activate macrophages and act as strong
chemo-attractants for circulating monocytes. Endothelins affect vascular
smooth muscle fibroblast proliferation; help regulate vascular, airway,
and intestinal smooth muscle tone; increase the activity of bone
alkaline phosphatase; stimulate release of atrial natriuretic peptide
(ANP) from atrial cardiocytes; inhibit the release of renin from
glomeruli and modulate norepinephrine at sympathetic nerve termini.
Opposing vasomotor effects are regulated through the binding of distinct
ET receptors for vasoconstriction and vasodilation. Endothelins have
been linked to atherosclerosis and various cardiovascular disease states.
ET-1 is important in congestive heart failure, renal failure,
pulmonary hypertension, hyperlipidemia and metastatic prostate cancer.
Big ET-1 is more stable than
ET-1 in plasma and may be useful in assessing ECE inhibitors. It
exerts similar effects in vivo, but has reduced vasoconstrictor
activity in vitro.
