Beta-catenin is a 781 amino acid adherens junction protein that plays an
important role in two unrelated cellular functions. Adherens junctions
are critical for establishment and maintenance of epithelial layers,
such as those found lining the surface of organs. Involved in both
cell-cell adhesion and in the wnt/wg signaling pathway, beta-catenin can
be found in the cytoplasm or the nucleus. In the cytoplasm, beta-catenin
is unstabilized by phosphorylation and association with E-cadherin. When
beta-catenin translocates to the nucleus, it has comparatively low
levels of phosphorylation. Wnt/wg signaling results in beta-catenin
accumulation and transcriptional activation of specific target genes
during development. Deregulation of beta-catenin signaling is an
important event in the evolution a number of malignancies, including
colon cancer, melanoma, ovarian cancer, and prostate cancer.

1. Pipet Standards and Samples in duplicate into the wells and incubate.
2. Aspirate and wash the plate.
3. Pipet the yellow Antibody into the wells and incubate.
4. Aspirate and wash the plate.
5. Pipet the blue Conjugate into the wells and incubtate.
6. Aspirate and wash the plate.
7. Pipet Substrate into the wells and incubate.
8. Pipet Stop solution and read plate on a plate reader at 450nm.
9. Calculate sample concentrations from the Standard Curve.
