Bax (Bcl-2 associated x protein) is an apoptosis regulating protein of
the Bcl-2 family. The ratio of Bax to Bcl-2 determines survival or death
following an apoptotic stimulus. Bax is implicated in many cancers,
uterine leiomyomas, and brain dysmorphology. Bax is a 21 kDa protein
that accelerates apoptosis by binding to, and antagonizing the death
repressor activity of Bcl-2 in vivo. Bax normally resides in the
cytoplasm, but following an apoptotic stimuli, Bax undergoes a
conformational change and translocates to mitochondrial membranes, where
it inserts and mediates the release of cytochrome c from the
intermembrane space into the cytosol, in this manner activating
caspase-3 and perpetuating the apoptotic cascade. Protein levels of Bax
and other Bcl-2 family members are altered in various human malignancies,
with changes in expression levels implicated in diseases involving
apoptosis such as cancer, autoimmune diseases and neurodegenerative
disorders.

1. Pipet Standards and Samples in duplicate into the wells and incubate.
2. Aspirate and wash the plate.
3. Pipet the yellow Antibody into the wells and incubate.
4. Aspirate and wash the plate.
5. Pipet the blue Conjugate into the wells and incubtate.
6. Aspirate and wash the plate.
7. Pipet Substrate into the wells and incubate.
8. Pipet Stop solution and read plate on a plate reader at 450nm.
9. Calculate sample concentrations from the Standard Curve.
