The Assay Designs' Correlate-EIATM kits are designed for the
quantitative measurement of 8-iso-Prostaglandin F2alpha
(8-iso-PGF2alpha) in alkaline hydrolyzed samples.
Isoprostanes are prostaglandin isomers that are formed in vivo by
cyclooxygenase (COX) dependent and non-COX oxidation of arachidonic
acid. 8-iso-PGF2alpha freely circulates throughout the body
and is eliminated in the urine. Esterfied isoprostane covalenty bound to
phospholipids and lipoproteins must be released by base hydrolysis
before being measured. 8-iso-PGF2alpha has been shown to be a
potent vasoconstrictor; a potential mediator of hepatorenal syndrome and
atherosclerosis and a mutagen in 3T3 cells and in vascular
smooth muscle cells. It has also been postulated to participate as a
pathophysiological mediator and is able to modify the fluidity and
integrity of membranes.
Assay Designs´ Direct 8-iso-PGF2alpha kit allows you to
carry out the hydrolysis and subsequent quantifications of your samples
for covalently bound isoprostane without solvent extractions, phase
separations or thin-layer chromatography. To measure free
8-iso-PGF2alpha, please refer to Assay Designs´ 8-iso-PGF2alpha
kit (Catalog Nos. 900-010 and 901-010.)

1. Pipet Standards and Samples in duplicate into the wells.
2. Pipet the blue Alkaline Phosphatase Conjugate followed by the yellow
Antibody into the wells and incubate.
3. Aspirate and wash the plate.
4. Pipet Substrate into the wells and incubate.
5. Pipet Stop Solution and read on a plate reader at 405 nm.
6. Calculate sample concentrations from Standard Curve.
