15-deoxy-Delta12, 14-Prostaglandin J2 (15-d-PGJ2)
is one of the ultimate dehydration products of PGD2. PGD2
is formed from PGH2 which itself is synthesized from
arachidonic acid by the enzyme prostaglandin synthetase. In the aqueous
solution PGD2 forms PGJ2. Albumin or other serum
proteins present in the sample will convert PGD2 to the
isomeric compound, Delta12-PGJ2. In males about
152 ng of Delta12-PGJ2 is excreted in urine per
day, and in women about half this amount is excreted. 15-d-PGJ2
has been shown to indicate adipogenesis and the gamma isoform of the
peroxisome proliferation activated receptor (PPAR-gamma).
1. Pipet Standards and Samples in duplicate into the wells.
2. Pipet the blue Alkaline Phosphatase Conjugate followed by the yellow
Antibody into the wells and incubate.
3. Aspirate and wash the plate.
4. Pipet Substrate into the wells and incubate.
5. Pipet Stop Solution and read on a plate reader at 405 nm.
6. Calculate sample concentrations from Standard Curve.
