15-(S)-hydroxyeicosateraenoic acid (15(S)-HETE) is a hydroxy derivative
of arachidonic acid made by 15-lipoxygenase contained in blood platelets,
neutrophils, eosinophils and reticulocytes. It is the most abundent
eicosanoid in human bronchi. It has been shown to be chemotactic and
chemokinetic for polymorphonuclear leukocytes and vascular smooth muscle
cells. 15(S)-HETE acts as a second messenger in angiotensin-II induced
aldosterone production. Evidence also suggests many other biological
activites for this compound including stimulating insulin secretion by
pancreatic tissue, inducing hyperphosphorylation of several major
cytoskeletal proteins, enhancing surface expression of integrin
receptors and platelet aggregation in tumor cells. It induces
immunosuppresion and inhibits ACTH corticosterone production.
1. Pipet Standards and Samples in duplicate into the wells.
2. Pipet the blue Alkaline Phosphatase Conjugate followed by the yellow
Antibody into the wells and incubate.
3. Aspirate and wash the plate.
4. Pipet Substrate into the wells and incubate.
5. Pipet Stop Solution and read on a plate reader at 405 nm.
6. Calculate sample concentrations from Standard Curve.
