Description:
Deoxyribonuclease I (DNase I) is an endonuclease that digests
single- and double-stranded DNA, chromatin and RNA:DNA
hybrids. It hydrolyzes phosphodiester bonds producing mono-
and oligodeoxyribonucleotides with 5'-phosphate and 3'-OH
groups. The enzymeactivity is strictly dependent on Ca2+
and is activated by Mg2+ and Mn2+ ions.
Applications:
- Degradation of DNA template in transcription reactions.
- Removal of contaminating genomic DNA form RNA samples.
- DNase I footprinting
- Nick translation
